Was Spain the first nation to find a sea route from Europe east Asia?

No, Spain was not the first nation to find a sea route from Europe to East Asia. The first were the Portuguese. Vasco da Gama, a Portuguese explorer, was the first to sail from Europe (Lisbon) to India (Calicut) in 1498, inaugurating an ocean route between Europe and Asia that would come to be known as the Cape Route.
